从古巴归来后，罗斯福再度进入纽约政界，1898年当选为州长。1900年大选，罗斯福当选副总统，1901年9月6日，总统麦金利被刺杀，不治身亡。罗斯福补位登上总统宝座，成为美国历史上最年轻的总统。他是唯一没有指着《圣经》宣誓就职的美国总统。1904年总统大选时，罗斯福在共和党内的主要对手病逝，遂使他轻而易举获得党内提名，并赢得大选，成功连任。
